چکیده

Plants have a highly sophisticated endomembrane system targeted by plant viruses for cell-to-cell movement. The movement protein (MP) of ourmia melon virus (OuMV) is delivered to plasmodesmata (PD) and forms tubules facilitate In our previous work, we identified the sites OuMV correct subcellular localization; however, pathways involved in PD-targeting MP are largely unclear, their identification was aim this work. study, demonstrate that localizes trans-Golgi network (TGN) but not multivesicular body/prevacuolar compartment or Golgi, carries two putative sorting motifs, tyrosine (Y) dileucine (LL) motif, near its N-terminus. Substitutions these motifs result loss infectivity Nicotiana benthamiana Arabidopsis. Live cell imaging GFP-labeled motif mutants reveals both required targeting PD efficient systemic infection show differences functionality. Moreover, co-immunoprecipitation assays coupled with mass spectrometry series host factors could interact link various pathways, particular vesicle trafficking membrane lipids. Our findings delineate route elucidate potentially recruited be modified impair infection.

Lipid-dependent protein sorting at the trans-Golgi network.

In eukaryotic cells, the trans-Golgi network serves as a sorting station for post-Golgi traffic. In addition to coat- and adaptor-mediated mechanisms, studies in mammalian epithelial cells and yeast have provided evidence for lipid-dependent protein sorting as a major delivery mechanism for cargo sorting to the cell surface. The mechanism for lipid-mediated sorting is the generation of raft pla...

Protein and lipid sorting from the trans-Golgi network to the plasma membrane in polarized cells.

The targeting of proteins and lipids to the cell surface domains of polarized cells is not a simple bulk flow process but requires sorting into distinct apical and basolateral pathways from the trans-Golgi network. Here, we describe the sorting determinants in the cargo molecules, the cellular sorting machineries responsible for the hierarchical read-out of the signals, and the mechanisms of ca...

The trans-Golgi network: a late secretory sorting station.

Proteins synthesized on membrane-bound ribosomes are transported through the Golgi apparatus and, on reaching the trans-Golgi network, are sorted for delivery to various cellular destinations. Sorting involves the assembly of cytosol-oriented coat structures which preferentially package cargo into vesicular transport intermediates. Recent studies have shed new light on both the molecular machin...

contribution of setting to the study of characters and the main idea in e. brontes: wuthering heights and t. hardys: return of the native.

this thesis aims to adduce an unmitigated and comprehensive explication concerning the relationship of three significant elements of fiction: setting, chracter and theme. my research is basically placed on two outstanding novels of the 19th century: emily brontes wuthering heights and thomas hardys return of the native. my endeavour lies in studying the correlation among the three above-mention...

ARF1 localizes to the golgi and the trans-golgi network.

The recruitment of coat proteins for transport vesicles (COPI-, COPII-, and clathrincoated) is mediated by the small GTPases of the ADP-ribosylation factor (ARF) family of which there are three SAR, 12 ARF, and six ARL genes in the Arabidopsis thaliana genome (Vernoud et al., 2003). These GTPases are themselves recruited by guanidine exchange factors located at the donor compartments, which con...
